چکیده

Bones from porphyric animals are treated first with 40% sodium hydroxide and subsequently with concentrated hydrochloric acid which dissolves the chalky residue from the alkaline digestion under violent foaming. The porphyrins are precipitated from the acid solution at pH ca. 3 together with calcium salts and subsequently washed and dissolved in about 50% acetic acid. The solution is then diluted with an approximately equal volume of water and the pH adjusted to ca. 3, after which the porphyrins are adsorbed on to a talc filter. After \vashing, the porphyrins are eluted with sulphuric acid-methanol and subsequently extracted with chloroform as their esters. Thin layer Chromatographie analysis of the ester crystals and mother liquors showed the presence of both uroand coproporphyrin and a series of intermediary porphyrins in bones from porphyric cattle and pigs.

Hepta-, hexa-, and pentacarboxylic porphyrins of porphyria cutanea tarda. II. Preparation of the porphyrins by stepwise decarboxylation ofuroporphyrins.

Hepta-, hexa-, and pentacarboxylic porphyrins have been detected in porphyric as well as normal urine samples. From the former they have been isolated and converted into pure methyl esters (1). For a better understanding of the chemistry of these porphyrins, pure uroporphyrin I and uroporphyrin III were separately subjected to decarboxylation under controlled conditions. The decarboxylation pro...

Porphyrins and Chlorins from a Congenital Porphyria Patient.

Analysis of porphyrins from the urine, blood, and feces of a congenital porphyria patient showed coproand uroporphyrins from the blood contained more than 95% of the Type I isomer, and those from stool and urine, about 90%. The intermediate hepta-, hexaand pentacarboxylic porphyrins from the urine contained 70% or less of the Type I isomer. The results are discussed and compared with those in p...

Toxicity of PBBs with special reference to porphyrinogenic action and spectral interaction with hepatic cytochrome P-450.

Some of the polyhalogenated aromatic compounds (PHAs) which are able to produce porphyria are presently known as environmental contaminants. Chronic exposure to PHAs causes hepatic porphyria in different species. Qualitatively PBBs act comparable to PHAs. An increase in accumulation of porphyrins caused by PHAs is not simply related to an increase of delta-ALAS activity in liver. Heme cannot ex...

Urinary total porphyrins by ion exchange analysis: reference values for the normal range and remarks on preformed prophyrins in acute porphyria urine.

Determination of total porphyrin with a rapid and east method (ion exchange; spectrophotometry) was performed on 57 morning urine samples from laboratory personnel, 59 arbitrary day urine samples from blood donors, and 90 24-hour urines from medical inpatients. The upper reference limit for morning urine was 0.32 mumol/l but even a value as high as 0.7 mumol/l was found. In the donor urines the...

Propofol as an i.v. anaesthetic induction agent in variegate porphyria.

The choice of an i.v. anaesthetic induction poses problems for the anaesthetist confronted with a patient with one of the acute porphyrias. We undertook a prospective clinical trial in 13 variegate porphyric subjects using propofol as an anaesthetic induction agent. Urinary porphyrin precursors and porphyrins were measured before operation and 1-5 days after operation. Stool and plasma porphyri...
